Design Notes
This sequence is a comprised of four different components: a constitutive promoter (pTet), lock (cis sequence), coding ccdB (without RBS), and a double terminator. The part sequences were placed one right after the other.
The pTet + cis sequence (BBa_J01060) was based on the composition part from the 2005 Berkeley iGEM team. The coding ccdB was based on the minimal ccdB gene (BBa_K414005) from 2010 Nevada iGEM team. We took the minimal gene and removed the RBS. The double terminator (BBa_B0015) was also from the iGEM registry.
